-Als voorloopige meêdeeling zij mij vergund eenige gedachten te
-vermelden, waarvan de uiting tot bespreking leiden kan; mij die ten
-nutte te maken, en zoo aan de eersten meer bepaaldheid en uitbreiding
-te geven is mijn doel.
-
-Daar het uitgangspunt voor de volgende beschouwingen gevonden is in de
-Chemie der Koolstofverbindingen, deel ik voorloopig slechts het daarop
-betrekking hebbende gedeelte meê:
-
-Meer en meer blijkt, dat de tegenwoordige constitutie-formules niet
-in staat zijn enkele isomerie gevallen te verklaren; wellicht is dit
-te wijten aan het ontbreken eener meer bepaalde uitspraak omtrent de
-werkelijke ligging der atomen.
-
-Neemt men aan, dat deze in een plat vlak zijn uitgebreid, zooals b.v.
-in Isobutylalcohol (fig. I), waar men de vier affiniteiten van elk
-koolstof-atoom door vier loodrecht op elkaâr in het plat vlak gelegen
-richtingen voorstelt, zoo komt men bij toepassing op de derivaten van
-METHAN (CH₄) (om van het eenvoudigste geval uit te gaan), tot
-het volgend aantal isomeren (de verschillende waterstof-atomen worden
-achtereenvolgens door univalente groepen R₁ R₂, enz. vervangen):
-
- Een voor CH₃ R₁ en voor CH (R₁)₃,
-
- Twee voor CH₂ (R₁)₂, (fig. II en III), voor CH₂ (R₁ R₂)
- en voor CH (R₁)₂ R₂,
-
- Drie voor CH (R₁ R₂ R₃) en voor C (R₁ R₂ R₃ R₄)
- (fig. IV, V en VI);
-
-welk aantal blijkbaar veel grooter is dan het tot nu toe bekende.
-
-Een tweede aanname brengt de theorie met de feiten in overeenstemming,
-n.l. die, de affiniteiten van het koolstof-atoom naar de hoekpunten
-eens tetraëders gericht te denken, waarvan dat atoom zelf het
-middelpunt is. Het aantal isomeren vervalt dan eenvoudig tot:
-
-Een voor CH₃ R₁, CH₂ (R₁)₂, CH₂ (R₁ R₂), CH (R₁)₃, en CH (R₁)₂ (R₂);
-doch twee voor CH (R₁ R₂ R₃), of algemeener voor C (R₁ R₂ R₃ R₄);
-want, denkt men zich in de lijn R₁ R₃ (fig. VII en VIII), het hoofd
-in R₁, ziende naar de lijn R₂ R₄, dan kan R₂ rechts (fig. VII) of
-links (fig. VIII) van den waarnemer zijn; met andere woorden: _Voor
-het geval de vier affiniteiten van een koolstof-atoom zijn verzadigd
-door vier onderling verschillende univalente groepen kunnen twee, en
-niet meer dan twee verschillende tetraëders worden verkregen, die
-elkaârs spiegelbeelden zijn, doch nooit zoo gedacht kunnen worden,
-dat ze elkaâr bedekken, d.i. men heeft te doen met twee isomere
-structuurformules in de ruimte._
-
-Volgens deze hypothese verkeeren de verbindingen C (R₁ R₂ R₃ R₄)
-in een ander geval dan C (R₁)₂ R₂ R₃, C (R₁)₃ R₂ of C (R₁)₄ welk
-onderscheid door de gebruikte voorstelling niet wordt uitgedrukt; dààr
-toch was tusschen C (R₁ R₂ R₃ R₄) en C (R₁)₂ R₂ R₃ even groot verschil
-als tusschen C (R₁)₂ R₂ R₃ en C (R₁)₃ R₂ of tusschen C (R₁)₃ R₂ en C
-(R₁)₄.
-
-Dit eerste resultaat der hypothese toetsend aan de feiten, meen ik
-er werkelijk in geslaagd te zijn te bewijzen, dat verbindingen,
-die zulk een koolstof-atoom bezitten (n.l. een gebonden aan vier
-onderling verschillende univalente groepen, in ’t vervolg asymmetrisch
-genoemd) ten opzichte van isomerie en andere eigenschappen afwijken,
-afwijkingen, die niet in de tot nu toe aangenomen constitutie-formules
-opgesloten liggen.
-

-
- EERSTE GEDEELTE.
-
-
-I. Verband tusschen de asymmetrische koolstof en het optisch actief
-vermogen.
-
-
-a. _Elke in oplossing draaiend op het polarisatie-vlak werkende
-koolstof-verbinding bezit een asymmetrisch koolstof-atoom._
-
-Het is slechts noodig het volgend lijstje van optisch actieve
-verbindingen na te gaan; in wier formules door =C= de
-asymmetrische koolstoffen aangegeven zijn, om zich van de juistheid
-dezer opmerking te overtuigen.
-
- Ethylidenmelkzuur: CH₃. =C.= H. OH. COOH.
- Asparaginezuur: COOH. =C.= H. NH₂(CH₂. COOH).
- Asparagine: COOH. =C.= H. NH₂. (CH₂. CONH₂).
- Appelzuur: COOH. =C.= OH. H. (CH₂. COOH).
- Glutarzuur: CH₂. OH. =C.= H. COOH. (CH₂. COOH).
- Wijnsteenzuur: COOH. =C.= H. OH. =C.= H. OH. COOH.
-
-Dextrose, Levulose, Gallactose, Maltose, Sorbin, Eucalin, enz.: CH₂.
-OH. =C.= H. OH. (C₄ H₇ O₄).
-
-Mannit, Quercit, Pinit: (C₄ H₉ O₄) =C.= H OH. CH₂ OH.
-
-Rietsuiker, Melksuiker, Melisitose, Melitose, Parasacharose en
-Trehalose; Zetmeel, Inuline, Glycogen, Dextrine en Arabine bevatten,
-als saamgestelde ethers van de boven gemelde verbindingen, ook de
-asymmetrische koolstof-atomen die daarin aanwezig zijn.
-
-Camfer, volgens KEKULÉ (fig. XII.)
-
-Borneol, volgens denzelfden (fig. XIII.)
-
-Camferzuur, volgens denzelfden, CO₂ H. =C.= H (C₈ H₁₄ O.)
-
-Terpentijnolie, waarschijnlijk geconstitueerd, zooals in fig. XIV; en
-Menthol, wellicht zooals in fig. XV is voorgesteld.
-
-Omtrent actieve alkaloiden, eiwitstoffen, enz. is nog te weinig van de
-constitutie bekend, om daaruit iets ten opzichte van verband tusschen
-structuur en actief vermogen te kunnen besluiten.
-
- * * * * *
-
-De eenige bepaalde uitzondering, die ik op dezen regel heb kunnen
-vinden, is de actieve propylalcohol van CHANCELL, doch,
-volgens een partikuliere meêdeeling van HENNINGER, is het
-betrekkelijk klein roteerend vermogen er van aan verontreiniging toe te
-schrijven.
-
-
-b. _Afgeleiden van optisch actieve verbindingen verliezen het draaiend
-vermogen, als de asymmetrie van alle koolstof-atomen vervalt; in ’t
-omgekeerde geval veelal niet._
-
-Enkele voorbeelden zijn hier voldoende:
-
-Inactief Malon-, Fumar-, en Maleïnzuur uit actief Appelzuur; Inactief
-Barnsteen- en Tartronzuur uit actief Wijnsteenzuur; Inactief Cymol uit
-actieve Camfer, enz.
-
-Daartegenover staan:
-
-Actief Appelzuur uit actief Wijnsteenzuur;
-
-Actief Wijnsteenzuur uit actieve Lactose;
-
-Actieve Glucosen uit actieve Glucosiden;
-
-Actief Nitromannit uit actief Mannit;
-
-Actief Camferzuur en Borneol uit actieve Camfer;
-
-Actieve zouten en ethers uit actieve zuren, enz.
-
-
-c. _Maakt men omgekeerd een lijstje van verbindingen, die een
-asymmetrisch koolstof-atoom bevatten, zoo is dadelijk te zien, dat in
-vrij veel gevallen het omgekeerde van_ (a) _niet doorgaat, d. i. niet
-elke verbinding met een zoodanig atoom schijnt op het gepolariseerde
-licht te werken;_ aan drie oorzaken kan dit worden toegeschreven:
-
-1. Daaraan, dat die verbindingen bestaan uit een inactief mengsel
-van twee gelijk maar tegengesteld optisch actieve isomeren, welke,
-door groote overeenkomst in hunne overige eigenschappen, moeielijk te
-scheiden, en tot nu toe niet gescheiden zijn.
-
-2. Aan het in ’t algemeen onvoldoend onderzoek van het
-rotatie-vermogen, hetzij wegens geringe oplosbaarheid, of wegens gering
-spec. draaiend vermogen van vele verbindingen, zooals b.v. met mannit
-het geval is.
-
-3. Daaraan, dat de voorwaarde „asymmetrische koolstof” voor optische
-activiteit onvoldoende is, en niet alleen het onderling verschillen der
-aan een koolstof-atoom hangende groepen, maar ook hun aard op zich zelf
-voorwaarde is.
-
-Wat ook van dat alles zij, toch leggen de opgemerkte feiten een’
-waarschijnlijkheidsband tusschen constitutie en actief vermogen, welke
-men, bij gebrek aan krachtiger argumenten, in de volgende gevallen
-bezigen kan:
-
-1. Een verbinding, die het gepolariseerde licht draait, bevat
-waarschijnlijk ook een asymmetrisch koolstof-atoom; hetgeen een middel
-geeft, bij eene voorloopig niet bepaalde constitutie aan zekere formule
-den voorkeur te geven, waardoor men o. a. een keus heeft bij de poging
-tot synthese.
-
-Voorbeeld. Draaiend amylal alcohol kan, met asymmetrische koolstof
-slechts
-
- CH₃
- CH. CH₂. OH
- C₂ H₅
-
-tot formule hebben; welke gedachte ook, doch op geheel anderen grond,
-door ERLENMEIJER is uitgesproken.
-
-2. Eene verbinding, welke tot nu geen physisch isomere heeft,
-die op gepolariseerd licht werkt, bevat met eenige mate van
-waarschijnlijkheid, geen asymmetrisch koolstof-atoom; ook dit kan op
-dezelfde wijze van dienst zijn, zoo b.v. kan Citroenzuur, in aanmerking
-nemende de vorming van Aconitzuur en Tricarballylzuur, slechts
-
- =C.= H. OH. COOH CH₂. COOH
- CH. COOH of C. OH. COOH
- CH₂. COOH CH₂. COOH
-
-
-tot constitutie hebben; het inactief zijn geeft aan de tweede formule
-eenigen voorkeur; de eerste toch bevat een asymmetrisch koolstof-atoom,
-waarom ik hoop genoemd zuur volgens FRANKLAND en
-DUPPA’s methode uit Oxalzure- en Jodazijnzure ether met behulp
-van Zink te verkrijgen.
-
-3. Eindelijk kan men met eenige mate van waarschijnlijkheid het
-grensgebied der draaiende verbindingen, d. i. de meest eenvoudige met
-actief vermogen aangeven:
-
-De eenvoudigste actieve eenatomige alcohol zal b.v. zijn:
-
- CH₃. CH. OH. CH₂. CH₃
-
-Het eenvoudigste actieve eenatomige zuur:
-
- CH₃. CH. COOH. CH₂. CH₃
-
-De eenvoudigste actieve tweeatomige alcohol:
-
- CH₃. CH. OH. CH₂. OH
-
-De eenvoudigste actieve verzadigde koolwaterstof:
-
- CH₃
- CH. C₃. H₇
- C₂ H₅
-
-De eenvoudigste actieve aromatische koolwaterstof:
-
- CH₃
- CH. C₆ H₅, enz.
- C₂ H₅
-
-tevens is het waarschijnlijk, dat enkele reeksen van actief vermogen
-zullen blijven uitgesloten, zoo b.v.:
-
- De normale koolwaterstoffen CH₃ (CH₂)ₙ CH₃
- De normale alcoholen CH₃ (CH₂)ₙ CH₂ OH
- De normale zuren CH₃ (CH₂)ₙ COOH, enz.
-
-merkwaardiger is het echter, dat, volgens de gemaakte opmerking, C H Br
-Cl I waarschijnlijk in twee isomeren zal kunnen gesplitst worden, die
-op het gepolariseerde licht werken.
-
-
-II. Verband tusschen de asymmetrische koolstof en het aantal isomeren.
-
-Zoo wellicht niet de asymmetrische koolstof elke verbinding,
-waarin ze voorkomt, optisch actief maakt, toch moet ze, volgens
-de grondhypothese, een isomerie veroorzaken, die zich op de eene
-of andere wijze uit, een isomerie die het door de tegenwoordige
-structuur-formules vooruitgeziene aantal, bij aanwezigheid van één
-asymmetrische koolstof, verdubbelt, en het bij toeneming vermeerdert
-bij aanwezigheid van meerderen.
-
-Werkelijk meen ik, dat er op verbindingen gewezen kan worden, die deze
-schijnbare anomalie vertoonen, welke WISLICENUS met den naam
-van geometrische isomerie bestempelde, wijzende op het onvoldoende
-der tegenwoordige voorstellingen, zonder evenwel eene zich beter
-aansluitende hypothese op te werpen.
-
-Daaronder meen ik te mogen noemen:
-
-De Ethylidenmelkzuren, welke één asymmetrisch koolstof-atoom hebben;
-
-De Wijnsteenzuren, Dibroom- en Isodibroom barnsteenzuur, Citra- Ita- en
-Mesabrompyrowijnzuur, Citra- Ita- en Mesa-malzuur, Mannit en isomeren,
-Dextrose en isomeren, wellicht ook Terpentijnolie, suikers, enz. met
-hunne isomeren, in allen waarvan meerdere asymmetrische koolstof-atomen
-tot de verhooging van het aantal isomeren samenwerken.
-
-
-
-
- TWEEDE GEDEELTE.
-
-
-Tot hiertoe is alleen de invloed na-gegaan der gestelde hypothese op
-verbindingen, waar de koolstof-atomen door één affiniteit aan elkaar
-gebonden zijn (daargelaten enkele aromatische lichamen); uit den aard
-der zaak sluit dus hier zich aan:
-
-_Invloed der nieuwe voorstellingswijze op verbindingen met
-dubbelgebonden koolstof-atomen._
-
-De voorstelling eener dubbelbinding wordt twee met een ribbe tegen
-elkaâr liggende tetraëders (fig. IX), waarin A en B de bindingen
-der twee koolstof-atomen, R₁, R₂, R₃ en R₄ de univalente groepen
-voorstellen, waardoor de overblijvende vrije affiniteiten der
-koolstof-atomen zijn verzadigd.
-
-Zijn R₁, R₂, R₃ en R₄ dezelfde groepen, dan is slechts één figuur
-denkbaar, eveneens als alleen R₁, en R₂ of R₃, en R₄ dezelfde zijn;
-_zoo echter tegelijk R₁ en R₂, benevens R₃ en R₄ verschillen, waarbij
-toch R₁ en R₃, R₂ en R₄ gelijk kunnen zijn, dan worden twee figuren
-denkbaar, voorgesteld in fig. IX en X, wier verschil veroorzaakt wordt
-door de relatieve ligging der groepen R₁ en R₂ ten opzichte van R₃
-en R₄; het verschillen dezer figuren, welke zich tot het aantal twee
-bepalen, voorspelt een geval van isomerie, hetwelk in de vroegere
-voorstellingswijze niet ligt opgesloten._
-
-Terugkeerend tot de feiten meen ik hiervan bij organische verbindingen
-gevallen te hebben aangetroffen:
-
-1. Maleïn- en Furmarzuur, ten opzichte waarvan alle verklaringen
-der isomerie schipbreuk hebben geleden (ik reken ook daartoe de
-aanname eener bivalente koolstof, daar deze alleen bij Kooloxyd
-en Carbylaminen, om bepaalde redenen, zonder verdubbeling van het
-molekuul, bestaan kan); werkelijk verkeeren deze zuren in het
-hier-boven beschreven geval: _Twee dubbel gebonden koolstof-atomen
-dragen elk twee onderling verschillende univalente groepen, H en
-COOH._
-
-2. Brom- en Isobrommaleïnzuur; de verklaring dezer isomerie is geheel
-dezelfde: men heeft slechts een H in het Fumar- en Maleïnzuur door een
-Br. te vervangen.
-
-3. Citra-, Ita- en Mesaconzuur. Bij aanname van
-
- CH₃. CH. COOH. CH₂. COOH
-
-voor pyrowijnsteenzuur blijven voor genoemde zuren slechts de formulen:
-
- CH₂=C. COOH. CH₂. COOH
- en CH₃. C. COOH=CH. COOH
-
-over; en, zoo niet de laatste, volgens mijn voorstelling, twee
-isomeren (waarschijnlijk Ita- en Citraconzuur) in zich sloot, zou geen
-ongedwongen verklaring te geven zijn.
-
-4. Vast en vloeibaar Crotonzuur. De constitutie van het vast crotonzuur
-is, volgens KEKULÉ, ontwijfelbaar:
-
- CH₃. CH.=CH. COOH
-
-voor het vloeibaar crotonzuur blijft dus, (alzoo werd er geredeneerd)
-niet anders dan:
-
- CH₂=CH. CH₂. COOH
-
-over, ten einde het niet identiek zijn te verklaren.
-
-Neemt men echter de volgende feiten, betrekking hebbende op dit zuur,
-in aanmerking:
-
-a. Met K O H gesmolten geeft het, volgens M. HEMILIAN,
-uitsluitend azijnzuur,
-
-b. Oxydatie-middelen zetten het, volgens denzelfden, in azijnzuur,
-oxalzuur, en, indirekt uit ozalzuur, koolzuur om,
-
-c. Bij 170°-180° gaat het, eveneens volgens HEMILIAN, in vast
-Crotonzuur over;
-
-zoo pleit niets vóór de formule CH₂=CH. CH₂. COOH, en alles voor een
-isomeer CH₃. CH=CH. COOH, geheel analoog aan het Fumar- en Maleïnzuur;
-werkelijk voldoet ook de formule CH₃ CH=CH. COOH, aan de eischen, door
-mijn hypothese voor de mogelijkheid van twee isomeren gesteld: twee
-dubbel gebonden koolstof-atomen, wier vrije affiniteiten elk door twee
-verschillende univalente groepen, in dit geval H en CH₃, H en COOH
-verzadigd zijn.
-
-5. GEUTHER’s Chloorcrotonzuur en Chloorisocrotonzuur, waarvan
-tot nu toe de isomerie werd uitgedrukt door:
-
- CH₂=CH. Cl. CH₂. COOH
- en CH₃. CCl=CH. COOH
-
-geven volgens FROELICH met Waterstof in statu nascenti de onder (4)
-behandelde zuren, waardoor beider constitutie
-
- CH₃. CH. CCl.=CH. COOH
-
-wordt, en dus hun isomerie-geval mijn hypothese sterkt.
-
-
-
-
- DERDE GEDEELTE.
-
-
-Ter behandeling blijven nu nog de drievoudig gebonden koolstof-atomen,
-zooals b.v. die in Acetyleen; de binding wordt hier voorgesteld
-door twee met drie hoekpunten, dus met een grondvlak in elkaâr
-liggende tetraëders (fig. XI): A C B is de drievoudige binding, R₁
-en R₂ zijn de univalente groepen, waardoor de twee overblijvende
-koolstof-affiniteiten verzadigd worden; de nieuwe hypothese geeft
-hier geen aanleiding tot afwijking van de tegenwoordig gevolgde
-beschouwingen.
-
- * * * * *
-
-Tot besluit meen ik te mogen opmerken, dat
-
-1. De nieuwe hypothese niets onverklaard laat, wat door de
-tegenwoordige opvatting duidelijk is.
-
-2. Enkele eigenschappen en isomeriën, door de gevolgde voorstelling
-niet opgehelderd, krijgen door deze beschouwingen eenig licht.
-
-3. Eindelijk sluit zich mijne opmerking omtrent in oplossing actieve
-verbindingen, d. i. actieve molekulen, aan RAMMELSBERG’s beschouwingen
-omtrent actieve kristallen aan; deze toch beweert, voortbouwende op
-HERSCHELL’s en PASTEUR’s waarnemingen, dat de eigenschap in vasten
-toestand op het polarisatie-vlak te werken (d. i. dus zoowel het actief
-zijn van kristallen uit inactieve molekulen, als het inactief zijn
-van kristallen uit actieve molekulen) samengaat met het optreden van
-twee niet volkomen overeenstemmende kristalvormen, die evenwel elkaârs
-spiegelbeelden zijn. Duidelijk is het, dat men hier te doen heeft met
-een rangschikking van molekulen in het actieve kristal, geheel analoog
-aan de asymmetrische rangschikking der atoomgroepen in het actieve
-molekuul volgens mijn hypothese; eene rangschikking, die veroorzaakt,
-dat, zoowel de door RAMMELSBERG opgesomde actieve kristallen, als de in
-fig. VII en VIII algemeen voorgestelde actieve molekulen, geen vlak van
-symmetrie bezitten.
